A meta-analysis of 12,339 borderline personality disorder cases and 1,041,717 controls found 11 independent genomic loci and 9 risk genes, the first genome-wide significant hits reported for a personality disorder.
The study estimated BPD SNP heritability at 17.3% on the liability scale, while polygenic scores explained 4.6% of variance and raised case odds 6.61-fold for the top versus bottom decile.
Genetic overlap was strongest with PTSD, depression and ADHD, with correlations of 0.77, 0.74 and 0.67, and also extended to antisocial behavior, suicide, self-harm and trauma-related traits.
