At least 55 people were killed in Kaung Tat village in Shan State, with dozens more wounded and rescuers still pulling victims from rubble.
The TNLA, which controls the area near the Chinese border, said explosives used for mining and quarrying detonated in the rebel-held village.
Residents described devastation across almost an entire neighborhood, saying children were among the dead and hundreds of homes were damaged.
One survivor said many initially feared an air strike, then questioned why explosive materials had been allowed so close to homes.
The toll rose from earlier reports of 46 deaths, underscoring the scale of the disaster in a region already shaped by fighting between the TNLA and Myanmar's junta.
