Jemele Rhone, 30, was charged with murder and Deiryen Dyce, 32, with assisting an offender over the fatal shooting of Shanice Brookes in Sheffield.
Brookes, 30, was found with serious injuries outside One Four One on West Street at about 02:45 BST on Monday and later died in hospital.
South Yorkshire Police said the mother of one and charity worker was an innocent bystander on a bank holiday night out when she was killed.
Both defendants are from Sheffield, and the charges mark the first major step in the investigation into the city-centre shooting.
