Fifteen fire engines and about 100 firefighters were deployed to a three-storey supermarket building with flats above on Golders Green Road after the first of 56 calls came in shortly after 06:45 BST.
London Fire Brigade said the fire was producing a significant amount of smoke, prompting advice for nearby residents to keep windows and doors shut while crews from Willesden, Finchley and West Hampstead worked at the scene.
Met Police said no injuries had been reported and officers were helping evacuate people from the building.
Road closures were imposed around Golders Green Road near Beverley Gardens, and authorities said the cause of the fire was still unknown.
