  • Scientists have identified early dopamine dysfunction in the entorhinal cortex as a key driver of memory loss in Alzheimer’s disease.
  • Studies in Alzheimer’s mouse models showed dopamine levels dropped by over 80%, impairing memory formation, but memory was restored using Levodopa or optogenetic stimulation.
  • This discovery highlights dopamine’s crucial role in memory and suggests existing Parkinson’s drugs could be repurposed for early Alzheimer’s treatment.
This breakthrough worked in mice, but can Levodopa truly reverse memory loss in humans?

Restoring Dopaminergic and GABAergic Balance: Emerging Therapies Targeting Memory Circuits in Alzheimer's Disease

Overview

Damona Pharmaceuticals is developing DPX-101, an oral drug that selectively enhances the α5 subunit of GABA-A receptors to restore cognitive function. Preclinical studies showed DPX-101 reverses memory deficits in Alzheimer's and other brain disorders without side effects, leading to FDA clearance and a Phase 1 trial starting in 2025. Meanwhile, research from UC Irvine revealed that dopamine levels in the entorhinal cortex drop drastically in Alzheimer's, causing memory failure, which can be rescued by Levodopa or optogenetic stimulation. These findings highlight dopamine depletion’s role in disrupting GABAergic neurons and neural circuits, supporting a promising combination therapy of DPX-101 with dopamine restoration to rebalance brain circuits and improve cognition.
